Popis: SG-Glutamic-C™ is a serine endopeptidase, from<i> S. aureus V8</i>, that is highly specific for the cleavage of peptide bonds at the carboxy side of either aspartic or glutamic acid, depending on the buffer used. In Tris-HCl buffer, in particular in the absence of phosphate ions, the enzyme is specific for the glutamyl site. Recommended buffers for fragmentation of proteins using this enzyme are 50 mM Tris-HCI, pH 8,0 or bicarbonate buffer. Highly purified preparations of SG-Glutamic-C™ are chemically modified making the enzyme both resistant to autolysis and stabilises its enzymatic activity.

Popis: One of the most fascinating cell envelope structures in prokaryotic organisms is two-dimensional arrays of protein or glycoprotein subunits, termed S-layers (Surface Layers). They are composed of numerous identical subunits forming a symmetric, porous, lattice-like layer that completely covers the cell surface. The subunits are held together and attached to cell wall carbohydrates by non-covalent interactions, and they spontaneously reassemble in vitro by an entropy-driven process. In lactobacilli, S-layer proteins have been detected on many but not all species. Lactobacillus S-layer proteins differ from those of other bacteria in their smaller size and high predicted pI. The positive charge in Lactobacillus S-layer proteins is concentrated in the more conserved cell wall binding domain, which can be either N- or C-terminal depending on the species. The more variable domain is responsible for the self-assembly of the monomers to a periodic structure. The biological functions of Lactobacillus S-layer proteins are poorly understood, but in some species S-layer proteins mediate bacterial adherence to host cells or extracellular matrix proteins or have protective or enzymatic functions. Lactobacillus S-layer proteins show potential for use as antigen carriers in live oral vaccine design because of their adhesive and immunomodulatory properties and the general non-pathogenicity of the species.

Popis: The procedure for determining dietary fibre exposes the sample to a series of enzymatic digestions that simulate the real digestive process which takes place in the human or animal digestive tract, calculating the undigested residue remaining at the end of the analysis. Generally dietary fibre analysis is carried out on foods intended for human consumption whereas raw fibre analysis is carried out on animal feeds or on raw materials of vegetable origin, eg. cereals. The GDE performs enzymatic digestion, a delicate phase where samples are immersed in a thermostatic water bath and stirred. Continuous and constant sample mixing is necessary in order to prevent the sample from overheating. The unit consists of an immersion thermostat, a transparent polycarbonate tank and a VELP 6-place magnetic stirrer to ensure excellent thermoregulation and precision. The use of deionised water is recommended as it eliminates any possible cause of limescale.
