Popis: Interleukin 31 (IL-31) is a recently discovered T-cell cytokine closely related to IL-6 type cytokines and is preferentially produced by T helper type 2 cells. IL-31 activity is mediated through the ligand-induced oligomerization of a dimeric receptor complex containing IL-31 receptor A and oncostatin M receptor. In response to IL-31 binding, these proteins activate the JAK/STAT and the AKT signaling pathways. RNA levels of IL-31 receptor A and oncostatin M receptor are induced in activated monocytes but are expressed constitutively in epithelial cells. IL-31, when overexpressed in transgenic mice, results in the development of pruritis, alopecia and skin lesions, and in humans may result in atopic dermatitis, suggesting that IL-31 may represent a novel target for antipruritic drug development.

Popis: Alpha-lactalbumin is the B protein of lactose synthetase secreted by the mammary epithelial cells. It is a potent Ca2+-elevating and apoptosis-inducing agent with broad, yet selective, cytotoxic activity. Multimeric lactalbumin has been shown to kill all transformed, embryonic and lymphoid cells tested, but not mature epithelial elements. This suggests that milk contributes to mucosal immunity not only by furnishing antimicrobial molecules but also by policing the function of lymphocytes and epithelium. ?lactalbumin may be helpful in discovering the site of origin of metastatic breast tumours. Human lactalbumin contains 123 amino acid residues. Comparison of the 5' flanking sequences of the two Alpha-lactalbumin genes with those of five casein genes reveals the presence of a highly conserved region extending from position -140 to -110 in all seven sequences examined, suggesting a possible regulatory role in the hormonal control or tissue-specific expression of milk protein genes in the mammary gland.

Popis: p75NTR (CD271) was originally discovered as a low affinity nerve growth factor receptor (NGFR). Later it was found that it was the receptor for all neurotrophins, including NGF, BDNF, NT3 and NT4/5. It mediates signals of neurotrophins for neuronal survival, apoptosis, neurite outgrowth and synaptic plasticity. Recently, it has been revealed that p75NTR not only acts as the receptor for neurotrophins but also the receptor for many other pathological ligands such as prions, rabies virus and amyloid beta. p75NTR also acts as a co-receptor for NOGO which mediates inhibitory signals of myelin associated protein. p75NTR is highly expressed in a number of non-neuronal and neuronal cells including motor neurons during development and also in damaged neurons. Recent research proposes the extracellular domain of p75NTR as a biomarker for monitoring the progression of motor neuron disease (MND), also known as Amyotrophic Lateral Sclerosis (ALS) or Lou Gehrig's Disease.

Decreased fibulin-5 may contribute to the pathogenesis of aortic dissection by impairing elastic fiber assembly. Betatrophin may also function in inhibition of lipase activity and on serum triglyceride regulation. Betatrophin is expressed in the liver and in white and brown adipose tissue of mice. In humans, betatrophin is found to be predominantly expressed in the liver. Betatrophin levels are reduced by fasting and are elevated upon insulin resistance and during pregnancy. Betatrophin, according to preliminary data could bind to the macrophage receptor Marco and also to RTN4R, a neuronal receptor.
